15 生物化学分离纯化    (部分)

生物化学本身有科学问题,也含可以推动其他学科的技术。

生物化学分析生物体内的分子,尚未结束。生物化学分离纯化生物体系的分子,分析其化学特性和生物学功能,长期以来颠扑不破地对生命科学有重要贡献。在生物问题的分子水平理解完成之前,在未来很长时期,分析和纯化仍将对生物学研究很重要。

生化纯化的关键是:有检测生物学活性的方法,获得足量的含有活性的组织、细胞来源,有合适的分离纯化技术和方法。分离纯化到分子,推动生物学问题的理解。

纯化依赖的典型检测方式是化学反应,也可以是细胞生物学现象,还可以是病理症状。纯化可以很快,也可以很慢。设计的巧妙和长期的坚持,各有所用。

15.1生物化学分离纯化的重要性

生化提纯当然首先应用于生物化学本身的问题,如酶促反应、代谢、信号转导,但也应用于很多其他学科,例如分离到激素,揭示了内分泌的基础。

脑和神经的功能复杂,但其研究在有些方面也得益于生化分离提纯。十九世纪,德国海德堡的Willy Kühne分离视紫红质是视觉研究的里程碑(Kühne,1879,1882),1950至1960年代,美国华盛顿大学Victor Hamburger、Rita Levi-Montalcini、Stanley Cohen分离提纯神经生长因子是发育神经生物学的里程碑、也开启生长因子的研究(Levi-Montalcini and Hamburger,1951,1953;Cohen, Levi-Montalcini and Hamburger,1954;Cohen,1960),并因为带动表皮生长因子的研究而间接促进了癌症研究(Cohen,1962)。分离纯化垂体激素是和神经内分泌的重要工作(Guillemin, 1978; Schally)。分离纯化神经肽是神经传递的重要工作(Euler and Gaddum,1931;Chang and Leeman,1970;Hughes et al., 1975)。分离纯化突触囊泡的膜蛋白,为理解神经递质释放提供了必要基础(Südhof and Jahn,1991)。神经纤维连接的重要问之一是轴突导向,理解其分子机理的途径之一也是分离提纯(Walter et al., 1987;Luo et al., 1993,1995; Drescher et al., 1995)。

本章先以轴浆转运的生化研究为例,了解分离纯化的基本要素和意义。再以Prion(沅蛋白)的发现为例,看长期坚持分离纯化带来意想不到的概念。

15.2-15.5轴浆转运的分子机理

15.6 羊的痒病及其传染

动物的疾病,或人的罕见病,研究有什么意义?“值得”坚持不懈的进行研究吗?还要经常与人发生争论?与众不同而被嗤之以鼻,出现苗头又担心竞争?

一项研究,如果几天做实验可以,如果要等几十天、上百天才知道结果,而且还要反复很多次,那还做吗?

在1732年就记载有绵羊的瘙痒病(McGowan,1914),后来有过多种名称,最后定为scrapie。1913年被认为是传染病(Stockman,1913),提出致病原因可能是寄生虫感染(McGowan,1914)或遗传因素(Stockman,1926)。文献记载此病曾在欧洲、美洲、亚洲、大洋洲、非洲都曾多次爆发(Detwiler,1991)。德国科学家最初未见行为或病理变化(Cassirer,1898),但法国科学家则发现在脊髓的前角细胞有空泡(Besnoit and Morel,1898a,1898b)。英国农业部首席兽医Stewart Stockman(1869-1926)在多个部位观察到神经细胞的空泡样病变(Stockman,1926)。冰岛科学家提出绵羊的几种病(包括痒病)是由慢病毒造成的进行性脑炎(Sigurdsson,1954)。

法国科学家发现,将痒绵羊的脑匀浆注射到正常山羊脑内,可以导致后者发生痒病(Cuillé and Chelle,1936),揭示其传染性。英国也发现,用患有痒病的绵羊脑组织制造的疫苗免疫动物,无心地将痒病传染给了其他绵羊或猪(Gordon,1946),从旁支持了其传染特征。介导传染的“病毒”可以通过滤膜,可以在0°到4°C存活很长时间,有时达两年,而通过脑注射引起疾病的潜伏期需要4、5个月(Wilson, Anderson and Smith,1950)。

到1960年代,研究者已观察到:绵羊传绵羊(Cuillé and Chelle, 1936; Gordon, 1946; Wilson, Anderson and Smith, 1950),绵羊传山羊(Cuillé and Chelle, 1939; Pattison, Gordon and Millson, 1959; Stamp et al., 1959),山羊传山羊(Pattison, Gordon and Millson, 1959),山羊传小鼠(Chandler, 1961; Zlotnik and Rennie,1962),山羊传大鼠和小鼠传大鼠(Chandler and Fisher, 1963),小鼠传金仓鼠(Zlotnik, 1963);绵羊传大鼠、小鼠和金仓鼠(Zlotnik and Rennie,1965),小鼠传绵羊(Zlotnik and Rennie,1965)。而且观察到,患痒病绵羊的多种组织,如脑、脾脏、淋巴结、脑脊液,都具有传染力(Stamp et al.,1959)。

但曾有英国科学家认为痒病不是环境因素造成,而是遗传病(Parry, 1960, 1962),并且是典型的孟德尔常染色体遗传(Draper and Parry, 1962)。痒病也曾被怀疑类似多发性硬化(Field, Miller and Russell,1962),但被后来研究所否定(Dick et al.,1965)。羊的痒病曾被认为是肌肉的疾病(Bosanquet, Daniel and Parry,1956;Parry,1957),但后来还是被明确为神经系统的退行性病变(Beck, Daniel and Parry,1964;Zlotnik and Rennie,1965)。

15.7 人的库鲁病及其传染

太平洋岛国巴布亚新几内亚曾为澳大利亚管辖。1950年代,澳大利亚巡警们多次注意到Fore部落有人发生不伴有发烧的颤抖、最后进展至死亡,当地人称“kuru”(Liberski et al.,2019)。片区医生Vincent Zigas(1920-1983)感兴趣研究,与来访的美国科学家D. Carleton Gajdusek(1923-2008)合作,他们描述了库鲁病的典型症状,发现家族聚集性(Zigas and Gajdusek,1957;Gajdusek and Zigas,1957)。家族性的发现导致有科学家提出单基因显性遗传的可能性(Bennett, Rhodes and Robson,1958)。病理切片发现神经细胞有空泡形成(Fowler and Robertson,1958)。

比较羊的痒病和人的库鲁病,在发病症状、发病进程、神经细胞空泡形成、神经退行性变等疾病特征上,它们都有相似性(Hadlow,1959)。

人类学家们发现当地习俗可能造成了此病的传染(Liberski et al.,2019)。在此基础上,Gajdusek等将病人脑匀浆注射到猩猩和猴,均引发库鲁病(Gajdusek, Gibbs and Alpers,1966),从而证明库鲁病是由脑内某种物质导致的传染病。因为感染到发病间隔数以月、甚至年计,Gajdusek提出病原体为“慢病毒”。

Gajdusek认为慢病毒不仅造成库鲁病,也造成多种其他疾病,当然包括羊的痒病,还包括人类的多发性硬化、帕金森病、侧枝硬化、阿兹海默病、亨廷顿舞蹈病等等(Gajdusek, 1977)。

15.8 痒病病原体化学本质的讨论

在早期的接种实验就知道,羊的痒病病原体很小,可以通过27微米直径的滤纸(Cuillé and Chelle, 1936; Stamp et al., 1959)。

痒病的病原体具有不同寻常的热稳定性,沸水煮30分钟,甚至更长的时间都不足以灭活(Wilson, Anderson and Smith, 1950,Stamp et al., 1959,Pattison and Millson, 1961;Chandler,1963;Hunter and Millson, 1964;Zlotnik and Rennie,1967)。对尿素和苯酚处理也有相当的抵抗性(Eklund, Hadlow and Kennedy,1963;Hunter et al., 1969)。0.35%的福尔马林三个月处理痒病病原体也不能灭活它(Gordon, 1946),甚至也在3%的福尔马林在37 oC处理13天后仍有活性(Stamp, 1962;Pattison, 1965)。

所以,虽然接种实验证明痒病是传染病,而其传染因子对高温和化学因素的抵抗性引起科学家们的困惑:不像一般的核酸蛋白复合体(Stamp et al., 1959)。

当时在英国工作的犹太物理学家Tikvah Alper (1909-1995)和同事,通过分析辐射对病原体灭活的特征,推测痒病的病原体分子量很小,如果是核酸应该少于800个碱基对(Alper, Haig and Clarke,1966)。病原体到底是核酸还是蛋白质,被作为问题而提出,并被怀疑可能是“自我复制的蛋白质”(Pattison and Jones,1967)。Alper等进一步在254nm波长进行高剂量的离子化辐射,几乎完全不能降低痒病病原体的传染性,因此不支持病原体依赖核酸而复制。但因为280至285nm波长的辐射对病原体的灭活作用也不强,她们未能证明是否就是蛋白质,因此讨论了病原体是多糖的可能性(Alperet al.,1967)。

本科念剑桥数学和生物化学、研究生专业为理论化学的英国科学家John Griffith(1928-1972),1967年就任伦敦大学Bedford学院数学系主任的第一个学术报告为“意识决策的神经基础”。他当年发表文章蛋白质自我复制的模型:如果是多聚体,一个亚基可以催化改变另外一个亚基的构型(Griffith,1967)。

痒病病原体影响细胞膜结构的可能性也被提出(Gibbons and Hunter,1967;Hunter et al., 1969),二十多年后还被Alper重新提出讨论(Alper,1992)。

15.9 探索分离纯化痒病的病原体

生物化学分类纯化羊痒病的病原体,是理解它的重要途径,事后看是“必由之路”。

分离纯化的努力始于1960年(Butler and Smith,1960)。最初,需要用绵羊作为检测的对象。从正常或感染痒病的绵羊在不同的pH条件提取脑匀浆、用丁醇等进行萃取,不同速度离心,得到上清和沉淀,注射到正常绵羊脑内,观察到:正常脑匀浆未引起30只受体绵羊的痒病,而痒病的脑匀浆(包括煮沸处理过的)都可以引起受体绵羊发生痒病(分别为3/15和1/30,发病/接种绵羊数),潜伏期为6到11个月(Mould and Smith,1962)。萃取方法不同、不同离心条件均能影响感染后的发病率(Le Q Darcel et al., 1963;Hunter and Millson,1964)。不同离心条件的实验也显示了其传染性与细胞核、线粒体、髓鞘没有相关性(Millson, Hunter and Kimberlin,1971)。

在这些努力的过程中,科学家们试过不同来源的病原体、不同动物用于检测。如曾经将绵羊的痒病脑注射到小鼠后,用小鼠脑匀浆作为病原体的来源,稀释后用多个剂量注射到正常小鼠作为检测动物(Siakotos Aet al., 1976),观察一年发病情况,并通过剂量推算半数致死率(LD50)。也比较过不同的组织,发现痒病感染的小鼠疾病进程较快(Siakotos, Raveed and Longa,1979)。

1960年代发现,将患痒病的小鼠匀浆注射到金仓鼠脑后,可以在4个月后发病(Zlotnik and Rennie,1965)。1970年代发现,在金仓鼠多次传代后,可以在六十多天就发病(Kimberlin and Walker,1977)。从而将需要等待6到11个月才能观察到发病,缩短到2个月,对于研究来说非常宝贵。

15.10 痒病病原体的分离纯化

犹太裔美国科学家Stanley Prusiner(1942-)念过医学院之后到NIH进行生物化学研究,然后到UCSF任住院医师。1972年,他看过的一位六十多岁的病人,因患罕见的神经退行性疾病(Creutzfeldt-Jakob病,CJD)而丧失记忆、几个月后死亡,两年后其脑匀浆注射给猴引起CJD样病变(Prusiner,2014)。

1974年,Prusiner成为UCSF神经内科助理教授,原来研究生物化学、已经发表过二十多篇论文的Prusiner决定研究羊的痒病。高年资同事普遍认为他不识时务。当时有笑话称痒病的假设多于研究痒病的科学家,因此是一个理论上充满含糊不清和争议的领域。他原先研究的脑内谷氨酸和谷胺酰胺是脑内富含的分子,而他转而研究在人里面类似的、也属于罕见病的兽医问题,这对于一个需要担心如何晋升终身教职的助理教授来说,是一种具有很大风险的职业选择。如果按当时用小鼠做实验的话,估计一年需要7万多小鼠,而观察期要一年,初算年度经费两百万美元,连续十年,如果不是绝对不可能,也是极为困难。

Prusiner决定集中全力研究痒病,系统地分离纯化其病原体。在Prusiner之前,已经有科学家在十几年前就开始分离纯化痒病的病原体,凭什么前人没有做到的,Prusiner可以做到?他研究动物的经验显然不仅不如其他人,而且他立即遇到能否获得足够的养传染病的动物空间问题。他获得的实验室空间,碰巧是爱因斯坦的媳妇Elizabeth Roboz-Einstein(1904-1995)退休空出来,但养动物的空间则费了很大周折,而且以后还几次有问题。他最初找蒙大拿州的痒病科学家William Hadlow(1921-2015)合作,在旧金山实验室分离纯化,然后送到蒙大拿州请合作者实验室用动物检测病原体传染活性。

他以自己的生化特长,认为以前做的不够系统。他仔细探索分离纯化方法,还专门为用固定角度转子的离心速度和方法发表文章(Prusiner,1978),并以分析性差速离心探索痒病病原体“沉淀特征”为其研究痒病的起点(Prusiner, Hadlow, Eklund and Race,1977)。他们开始用的痒病病原体来源是小鼠的脾脏,因为Hadlow和其他实验室以前发现病羊的脑匀浆注射到小鼠后,在脾脏中的痒病传染性升高最快(Eklund,Kennedy and Hadlow,1967;Hunter, Kimberlin and Millson,1972)。每次分离的组分稀释几个剂量,注射到正常小鼠,12个月每周观察临床症状(运动迟缓、尾巴弹性、体表粗糙,偶尔解剖脑和脊髓通过病理验证临床诊断)。最初拿到20倍的富集(Prusiner et al.,1978),并摸索了不同种类和浓度的去垢剂、阴离子、核酸酶、蛋白酶等如何影响分离纯化(Prusiner et al.,1980a)。他们意识到需要改用仓鼠作为检测的动物,而且只看注射后到发病之间的潜伏期长短,不用同一个组分多个剂量来确定量效关系,因为潜伏期就于量效关系直接相关,这样可以极大地节省实验时间和实验动物(Prusiner et al.,1980b)。

痒病感染金仓鼠是以前发现的(Zlotnik and Rennie,1965),其中包括长期看不上Prusiner的科学家(Marsh and Kimberlin,1975)。他们发现在金仓鼠传几代之后,潜伏期缩短到60-70天(Kimberlin and Walker,1977)。

但是,Prusiner再次遇到困难。蒙大拿的科学家不再与Prusiner合作,除非他搬过去。他只好在旧金山到处找养传染病动物的空间(Prusiner,2014)。好不容易找到后,他用金仓鼠做感染对象。虽然养金仓鼠的价格是小鼠价格的五倍,但用金仓鼠的潜伏期做检测指标,每个样本只要4只、而不是60只小鼠的,观察期也从小鼠的一年减为70天。改进方法后,他们进一步分离纯化并分析其组分。他们发现用RNA和DNA的水解酶都不能降低提取物的感染性,而用蛋白酶K可以部分降低感染性,用化学修饰蛋白质的焦碳酸二乙酯(Et2PC)可以降低感染性(Prusiner et al.,1981)。而用还原剂羟胺后,可以逆转Et2PC降低的感染性(McKinley, Masiarz and Prusiner,1981),因此支持蛋白质是病原体的重要成分。



他们也换了病原体来源。初期用金仓鼠的脾脏是因为感染后,脾脏的感染性上升比较快,但比较下来感染后金仓鼠脑中含的病原体的总量是脾脏的百倍,只是在分离纯化时先需要去除脑中的脂肪(Prusiner et al.,1982a)。

1982年,Prusiner给《科学》写了一篇独特的有趣文章,明确提出“新的蛋白质颗粒导致痒病”(Prusiner,1982)。这篇综述文章因其结论超前结果而被广为诟病,它提出导致痒病的病原体为prion。它提出人的CJD不仅可以传给猴和猩猩,也可以传给山羊和绵羊,而后者有可以传给猴和猩猩,也许可以传染给人?它总结了支持痒病病原体是蛋白质的结果(对蛋白水解酶的敏感性、化学修饰和逆转、对一部分去垢剂的敏感性,对一部分离子和尿素的敏感性,对甲醇乙醇等有机溶剂的抵抗而对苯酚的敏感)。它总结了不支持核酸是病原体的结果(对氢离子抵抗、对氢氧根敏感、对RNA酶和DNA酶抵抗、对254 nm波长紫外辐射的抵抗、对二价阳离子锌的抵抗、对羟胺的抵抗等)。其中,254 nm波长紫外辐射对多种噬菌体、多种病毒、类病毒都可以灭活,而痒病病原体的抵抗力远高于动物病毒和噬菌体。它的分子量大约50kD。Prusiner提出:要么病原体含核酸,编码prion蛋白质或激活宿主编码prion蛋白质的基因;要么病原体不含核酸,由蛋白质激活宿主编码prion的基因,或者蛋白质通过反向翻译编码自己的复制、或通过蛋白质指导的蛋白质合成编码自己的复制。

在同年,但以上文章发表之后,Prusiner实验室再在《科学》发表其分离纯化的结果(Bolton, McKinley and Prusiner,1982)。他们用碘的放射性同位素(125I)在感染痒病的金仓鼠脑中找到一个分子量在27到30kD的蛋白质,在正常金仓鼠脑中没有见到。有趣的是,这个蛋白质对蛋白酶K有较高的抗性。作者解释这是相对的,与病原体一样,在37oC用蛋白酶K处理三个小时以上可以灭活病原体、也可以降解这一蛋白质,抵抗力是相对的。他们用14C标记的实验,需要曝光720小时,也可以看到相应分子量的蛋白质(Prusiner et al.,1982b)。病羊脑中提取的prion蛋白(PrP)对蛋白酶K的敏感性低于正常羊脑的PrP,感染性与对蛋白酶K的抗性相关(McKinley, Bolton and Prusiner,1983)。在纯化的样本中,观察到短状的淀粉样纤维(rod,杆):直径10到20nm、长度100-200nm(Prusiner et al.,1983)。

Prusiner实验室进一步坚持,并大规模分离纯化,从九百到一千只感染了70到75天之后的金仓鼠脑,提取痒病的PrP,到1984年获得足够的大量、并且足够纯度的蛋白,与加州理工学院Lee Hood(1938-)实验室合作,用后者的蛋白质测序特长,得以分析PrP蛋白质的部分氨基酸序列(Prusiner et al.,1984)。

15.11 找到PrP基因:Prn

Prusiner与瑞士的Charles Weissmann(1931-)合作,找到了编码PrP的基因,他们称为Prnp。他们根据PrP蛋白质的部分序列设计核酸探针,用感染了痒病的金仓鼠的脑子制备cDNA文库。金仓鼠一般接种患病脑匀浆60到65天之后开始有症状,两周后死亡。他们用的是接种35天后的金仓鼠脑。用这个方法,他们找到了编码PrP蛋白质的基因Prnp(Oeschet al.,1985)。

找到Prnp的cDNA后,用它制备探针发现:在接种后的金仓鼠,无论是24小时,还是20天、40天、60天,PrP基因表达都一样。而且与正常金仓鼠也毫无差别(Oeschet al.,1985)。

那么,这怎么可能是致病的病原体?

一个竞争实验室也克隆了Prnp,也发现无论是金仓鼠还是小鼠,无论是脑还是肝、脾,Prnp基因的表达都不因为感染痒病而有变化,他们的结论自然就是是:PrP不可能是痒病的病原体(Chesebroet al.,1985)。

Weissmann和Prusiner进一步证明:痒病(Sc)和正常细胞(C)的PrP都是由同一个基因所编码,而且Prnp基因就在正常细胞的染色体上(Basleret al.,1986)。

此时,绝大多数人认为PrP致病的假说被颠覆,外界一片哗然,合作者Weissmann也不相信。Prusiner自己实验室做过PrP重要工作的博士后也不信,不仅不信还被反对他的人挖走(Prusiner,2014)。只有极少数的人认为Prusiner的研究不是笑话,而继续支持。绝大多数人认为致病因子应该还是核酸,只是Prusiner没有分离纯化到,而错误地纯化了一个蛋白质PrP,以自己名字命名一个没有证明功能的蛋白质。

15.12 两型PrP蛋白质

Prusiner坚持认为自己分离纯化是正确的,并接受所有已有结果,只是认为解释应该不在核酸,而在蛋白质。

他带领实验室其他人进一步研究PrP。

他们分析正常的PrP(PrPC)与致痒病的PrP(PrPSc)有同有异(Meyer et al.,1986):

前者存在于正常脑中、由细胞染色体的基因编码、mRNA长度2.1kb、蛋白质水平在正常与感染脑中不变、浓度低于1微克/每克脑组织、分离的病原体中没有、定位于在细胞膜上、细胞外没有、溶于去垢剂、容易被蛋白水解酶所降解;

后者不存在于正常脑中、也由细胞染色体的基因编码、mRNA长度也是2.1kb、而在感染的脑中增加、浓度约为10微克/每克脑中、每ID50感染病原体中含104分子、也定位于细胞膜上、在细胞外称淀粉样丝状斑块、去垢剂促淀粉样杆形成、蛋白水解酶导致它变成PrP 27-30。

15.13 Prnp与痒病感染

检验PrP功能方式之一是看Prnp基因型与痒病感染是否相关。

早期的研究发现,小鼠的遗传背景显著地影响痒病的潜伏期(Dickinson and Mackay 1964;Dickinson,Meikle and Fraser 1968),这与Prnp基因有什么关系呢?Prusiner等比较两种小鼠,它们感染痒病后发病的潜伏期差别很大,一种是110天左右、另一种是250天左右,并且这种潜伏期的差异可以稳定地遗传。Prusiner发现两种小鼠的Prnp基因不同,在小鼠交配的遗传实验中观察到基因差别与潜伏期相关(Carlsonet al.,1986)。进一步分析发现Prnp基因的具体碱基变化(第108密码子和189密码子)的差别与感染潜伏期相关(Westawayet al.,1987)。另外一种以前研究更长时间的小鼠,其潜伏期的差别也可以由Prnp基因的差异来解释(Hunter et al.,1987)。

在克隆了Prnp基因后,不仅可以观察小鼠自然出现的Prnp基因变异与痒病的关系,也可以通过转基因引入Prnp基因。已知金仓鼠感染痒病的潜伏期远低于小鼠的,Prusiner实验室把金仓鼠的Prnp引入小鼠。转了金仓鼠Prnp基因的小鼠感染痒病的潜伏期大大下降,病理切片也显示典型的淀粉样变(Scott et al.,1989)。进一步发现,小鼠表达转基因的PrP越多,感染的潜伏期越短(Prusiner et al.,1990)。转金仓鼠prnp基因的小鼠,只对金仓鼠来源的痒病脑匀浆的潜伏期缩短,而对来源于小鼠的痒病脑匀浆的潜伏期不变(Prusiner et al.,1990)。

15.14 Prnp与人类传染性神经退行性疾病

Gerstmann-Sträussler-Scheinker (GSS)是人类的一种家族遗传的神经退行性病变(Gerstmann , Sträussler and Scheinker,1936)。病人的脑匀浆注射到小鼠可以引起病变(Tateishi et al.,1979)。Prusiner实验室的Karen Hsiao(1955-)研究了两个家系,美国的有四代12人,对其中三个正常人、一个GSS患者的基因比较后发现病人的Prnp不同于其正常的亲戚。英国的家系4代54人,比较其中六位正常人和两位患者,观察到另外患者的Prnp基因变异(Hsiao et al.,1989)。这一差异是人的第102位脯氨酸的密码子突变编码亮氨酸,他们在小鼠(第101脯氨酸)制造同一位点的突变,观察到小鼠当突变基因高表达的时候,小鼠在166天左右自发出现神经退行性变,而且小鼠遗传发现只有得到突变的小鼠才自发神经退行性变、遗传了突变的绝大多数小鼠发病(Hsiao et al.,1990)。

Creutzfeldt-Jakob Disease (CJD)是人类一种可以家族遗传的神经退行性疾病(Creutzfeldt,1920;Jakob,1921)。CJD的脑匀浆注射到猩猩或豚鼠,都可以传染CJD(Gibbs et al.,1968;Manuelidis et al.,1976)。有趣的是,CJD病人也被发现含有prnp基因的突变(Owen et al.,1989;Goldgaber et al.,1989)。

人类的家族性致命失眠症(FFI),也是神经退行性变,也被发现含prnp基因突变(Medori et al.,1992)。后来也被证明可以传染(Tateishi et al.,1994)。

15.15 “全”prion

到1991年,可以确定PrP为痒病和人类神经退行性变的重要蛋白质。

但是,是异常结构的prion单独致病,还是它只是致病因素的一部分,是否还有其他因素?例如核酸?

总之,学界仍然难以接受蛋白质独自致病的理论,尤其是对痒病传播中所表现的种属屏障、株系,和株系突变等类似病毒传播的特征。

Prusiner的合作者Weissmann指出,有多种可能性可以解释prion如何导致疾病,并表现出非常强的种属屏障和株系特征。他提出,prion蛋白质只是致病源一部分,为apoprion,而与核酸(coprion)结合后成为完整的prion,为holoprion。传播和致病性由prion蛋白质(apoprion)所决定,不受核酸coprion影响。但与核酸coprion结合会影响prion蛋白的内在特性如蛋白构象,从而表现出种属屏障和株系特征(Weissmann,1991)。

15.16 Prnp基因敲除小鼠

Weissmann实验室在小鼠敲除了了prnp,没有观察到小鼠发育和生长与野生型的小鼠有何不同(Büeler et al.,1992)。

但是,prnp基因敲除的小鼠,对痒病脑匀浆接种具有完全的抗性,不会发病。而且如果再转基因导入金仓鼠的prnp基因,小鼠又可以被金仓鼠的痒病脑匀浆所感染(Büeler et al.,1993;Prusiner et al.,1993;Sailer et al.,1994)。

Prusiner实验室发现,如果将人类GSS突变基因引入小鼠,高水平表达转基因的小鼠自发地产生疾病,但低水平表达时,小鼠本身不发病。将高表达转基因的发病小鼠的脑匀浆注射给后者,可以显著提高发病率。高表达GSS突变但尚未发病的小鼠脑匀浆不能导致低表达GSS突变的小鼠发病。没有GSS突变的小鼠脑匀浆也不能引起低表达GSS突变小鼠发病(Hsiao et al.,1994)。

显然,prnp肯定对于痒病的传染是必需的。

15.17 PrP蛋白质的初步理解

在1980年代提出PrP蛋白质有致病的PrPSc和正常的PrPC两型后,在各种批评中,Prusiner继续研究。

人类正常细胞的Prnp基因编码PrP蛋白质,它的N端含信号肽。切断信号肽和C段部分序列之后的209个氨基酸是PrPSc和PrPC的序列,而分离纯化到的PrP 27-30是再切掉对蛋白酶敏感的N端后剩下的142氨基酸(Prusiner,1998)。

对PrPSc蛋白质的结构分析发现,它含约47%的b折叠结构,仅约14%的a螺旋结构(Caughey et al.,1992)。

要理解PrPSc和PrPC的差别,就不仅需要分离纯化和分析PrPSc,也需要分离纯化和分析PrPC。

分离纯化正常的PrPC蛋白质,也经过努力(Pan et al.,1992)。获得后,Prusiner实验室的Keh-Ming Pan等分析了其结构,并与PrPSc的结构进行比较。他们发现,PrPC的a螺旋占42%、b折叠占3%,而PrPSc的a螺旋占30%、b折叠占43%,所以提出正常PrPC变成致病的PrPSc主要原因是a螺旋降低、b折叠增加,也就是a螺旋构象改变成b折叠构象(Pan et al.,1993)。更好的结构分析方法也支持这种差别(Prusiner,1998)。

至此,PrP蛋白质的构象、致病和传染的模型确定:正常细胞编码PrPC蛋白质,病原体为PrPSc蛋白质。前者富含a螺旋,后者富含b折叠。当PrPSc感染时,相应的PrPC蛋白质被改变,其a螺旋改变为b折叠。这一过程可以复制,从而传染(Prusiner,1998)。

15.18 PrP蛋白质构象的间接遗传学支持

1996年,Prusiner实验室与合作者获得一个很有趣的遗传学结果,支持蛋白质影响蛋白质的构象。

CJD病人脑中对蛋白酶相对抵抗的PrPSc蛋白质在去除糖基化修饰后的大小为21kD,FFI病人脑中对蛋白酶相对抵抗的PrPSc蛋白质大小为19kD。它们分别接种转人PrP基因的小鼠后,小鼠患病,而小鼠脑中的对蛋白酶相对抵抗的PrPSc分别为21和19kD。也就是说,同样的小鼠,在接种不同来源的病人脑匀浆后,分别出现不同分子量的蛋白质。这种不同分子量是在蛋白酶水解后的结果。对这些结果的最简单推论是因为PrP构象不同,细胞内的不致病PrPC被来自人的PrPSc影响后,出现由感染的PrPSc构象所影响的鼠的PrP构象,所以对蛋白酶抵抗的区域大小有所不同。这种结果及其理解,支持蛋白质可以影响蛋白质构象,也就可能传播构象(Tellinget al.,1996)。

15.19 酵母的现象

如果有Prion这样以蛋白构象为基础的传播模式,那么,不可能只是为了传染病?

1971年,法国科学家在酵母中观察到一种遗传现象,不符合简单的孟德尔遗传学,曾认为也许是线粒体遗传,但也不符合线粒体遗传的预期(Lacroute,1971)。1994年,美国的Reed Wickner(1944-)用酵母中也有类似prion的传播现象,成功地解释了所谓两种基因的表型实际是由同一基因编码的、具有不同构象的蛋白质导致(Wickner,1994)。

美国芝加哥大学的Susan Lindquist(1949-2016)实验室更发现实际上在酵母中有很多遗传表型是由类似prion的蛋白质构象改变所致(Halfmann et al.,2012)。因为这些表型均为正常的酵母所有,符合她之前提出的蛋白构象也是遗传信息载体的理论(Halfmann and Lindquist,2010)。

15.20 体外复制

对体内现象的观察、分析和推理,可以有很好的见解。

但是,不需要核酸,只需要蛋白质改变其他蛋白质的构象,是非常特别的想法,需要严密的证明,只依赖体内实验及其分析不足以证明。

1994年,用金仓鼠的分离到的PrPSc蛋白质,在体外可以被一定浓度的盐酸胍变性、然后再稀释后能复性回具有蛋白酶K抗性的构象。而如果复性过程中与大肠杆菌表达的小鼠重组PrPC蛋白质孵育,观察到后者也能获得相对的蛋白酶抵抗性(Kociskoet al.,1994)。当同样的重组PrPC蛋白质与不同特性的PrPSc蛋白质共育后,获得的特性类似来源的PrPSc蛋白质(Bessen et al.,1995)。这些实验说明了PrPSc确实具有prion所需的模版功能,但能不能导致疾病呢?

以类似的方式得到的、具有PrPSc生化特征的PrP蛋白质,注射到动物却不能导致疾病(Hill, Antoniou and Collinge,1999)。瑞士科学家发明体外用发病的脑匀浆与正常脑匀浆混合之后扩增具有PrPSc生化特征蛋白质的方法(Saborio, Permanne and Soto,2001)。体外扩增的含PrPSc的产物具有感染性(Castilla et al.,2005)。

因为上述实验的产物是脑匀浆的混合物,它们的结果依然不足以完全证明蛋白质是致病源,公认的金标准是用重组PrP蛋白,在体外转变成具有蛋白酶K抗性的PrPSc构象,并在动物体内证明其致病性。

2004年,Prusiner实验室在大肠杆菌制造小鼠的PrP蛋白质89至230片段,当在体外多聚化成为淀粉样纤维后,注射到高表达编码小鼠89-231片段的转基因小鼠脑中,可以导致发病。但是潜伏期很长:380到660天。其脑中有蛋白酶抵抗的PrPSc蛋白质。从这样感染的小鼠中获得脑匀浆,注射到正常小鼠在150天导致发病,而如果是高表达全长PrP的转基因小鼠,接受注射后90天就发病。因此,Prusiner实验室认为这是证明PrP具有传染性的强有力证据(Legname et al.,2004)。但这一实验并没有平息争议(Couzin 2004),有科学家指出由于实验只在高表达PrP 89-231片段小鼠中成功(转基因表达量是小鼠内源PrP的16倍),而高表达PrP的转基因小鼠在老年会自发得病,注射的PrP淀粉样纤维可能只是压垮骆驼的最后一根稻草。

发病潜伏期太长和需要高表达转基因鼠的问题,于2010年为马继延实验室解决。他们在大肠杆菌表达小鼠PrP蛋白质之后,在体外摸索许多影响PrP构像的条件,他们最终获得一种条件可以将重组PrP在体外变构成具有PrPSc理化特征的构象,这样得到的PrP可以在130天导致野生型小鼠发病,与注射发病的脑匀浆完全一致(Wang et al.,2010)。

在酵母中,也做过类似的研究(King and Diaz-Avalos,2004;Tanaka et al.,2004)。

15.21 小结

对痒病的研究,从绵羊的罕见传染病,到人的罕见遗传病,从传染和遗传的混淆,到可能兼有,都是一般疾病研究过程。

而提出可能有不含核酸的病原体,是很特别的想法,但虽然有人提出过如此新颖的想法,如何研究它、是否持之以恒研究它,却并非热点。而且有些提出者自己也不坚信,后来放弃想法。

进行生物化学分离纯化,也已经开始,但研究传染病的人不一定很在行分离纯化。

一个初出茅庐的医生,不研究人的疾病,却研究绵羊的疾病,而且是罕见病,已经有人提出想法、至少十几年前就有人开始分离纯化病原体,领域还有获得诺贝尔奖并且对自己不友好的大人物,不顾研究经费和研究条件的艰辛,不怕自己职位尚未稳定的危险,开始系统地分离纯化病原体。在研究过程中遭遇各种冷嘲热讽,也有自己莽撞过早引来批评。

但是,用自己的特长,坚持不懈,严谨实验,不畏人言,以事实说话,推进了一个看似“罕见”,但在应用上的意义可以后面明显,在基础上需要很多不同方面的研究并入,在概念上非常新颖。

这与孟德尔有明显的不同,但也有相同。

科学,实验为主的生物学,选择正确的途径,按照实验观察到的事实而提出、或支持概念,新颖并非错误,虽然需要首先自己对自己有批判性,之后接受大家和时间的检验。

发现NGF最重要的三位科学家都长寿。犹太裔意大利生物学家蕾塔·列维-芒塔齐尼(Rita Levi-Montalcini,1909-2012)是第一位诺贝尔百岁寿星,享年103岁。她曾经的合作者、犹太德裔美国生物学家汉伯格(Viktor Hamburger,1900-2001)也逾百岁。他们是二十世纪后五十年最重要的两位发育神经生物学家,而前五十年是德国的Hans Spemann和美国的Ross G Harrison。美国犹太生化学家Stanley Cohen(1922-2020)在纯化NGF过程中起了主要作用,他还独立纯化了表皮生长因子(Epidermal Growth Factor,EGF),影响超出神经发育。

蕾塔·列维-芒塔齐尼作为孪生女出生于意大利Turin,读Turin大学医学院的同学还有两位诺贝尔奖得主:Renato Dulbecco(1914-2012)和犹太裔的Salvador Luria(1912-1991)。他们在Turin的导师都是Giuseppe Levi(1872-1965)。

二战期间,法西斯统治的意大利使犹太血统的蕾塔失学、失业,她就在家做实验。二战后的1947年,她到美国密苏里州的圣路易斯市,在华盛顿大学动物系(后称生物系)工作,做出重要发现,将近三十年后才回意大利。

汉伯格1920年代的研究生导师是德国胚胎学家、第一位发育生物学的诺贝尔奖得主Spemann。1932年汉伯格获洛克菲勒基金会资助到芝加哥大学合作研究,1933年德国通知他因为是犹太人而被除名,Spemann也来信告知爱莫能助。幸亏洛克菲勒基金会很快建立了紧急基金来支持被纳粹除名的科学家。汉伯格在芝加哥大学工作两年后,南迁密苏里州的圣路易斯市,加入华盛顿大学。

蕾塔一生相当专注于研究NGF,而汉伯格有多方面工作。他将德国的胚胎学传统带到美国,多年为神经胚胎学的领袖。他研究出色,还积极教学,写书,发挥了智力领导的作用,他的影响以及蕾塔·列维-芒塔齐尼的研究,使华盛顿大学长期为世界发育神经生物学的重镇。八十年代全世界学习发育神经生物学的教科书大多采用华盛顿大学Dale Purves和Jeff Lichtman编的《神经发育原理》。汉伯格除原创性研究,还做自己领域需要的描述性基础工作,例如他和Hamilton于1951年确定鸡胚分期的工作,虽非创造性研究,但非常实用,被引用已过万次。

15A1    细胞死亡和细胞生存

有很长时期,研究细胞的科学家只关注生长,而不注重死亡,以为死亡无需机制。

法国细胞学家Collin(1906)在题为“神经细胞发育的细胞学研究”一文中,已经注意到6日鸡胚中有细胞死亡,但并未意识到其重要性。1920到1930年代,德国海德堡大学Kallius指导的学生Ernst(1926)和Glücksmann(1930)研究高等动物中细胞死亡,他们进行了仔细观察,并将细胞死亡分类,他们明确地知道,在发育过程中,多处出现细胞死亡。

细胞死亡是否被调节?这个问题起源于研究肢体和神经的关系。功能上,肢体是神经所支配的靶组织,其中两种神经纤维,位于背根神经节的感觉神经细胞送出感觉神经纤维到达肢体、将肢体的感觉信息传入神经系统;位于脊髓腹角的运动神经细胞送出运动神经纤维到达肢体、将神经系统的指令传出到肢体指挥其运动。神经对肢体的支配发生在胚胎发育过程中,早期肢体刚开始发育时称为肢芽、或原基。

芝加哥大学Lillie实验室的研究生Shorey(1909)在鸡胚发现,电损毁前肢(翅膀)的芽盘后,支配它的感觉神经节和运动神经中心都减小。耶鲁大学的研究生Detwiler(1920)用蝾螈发现,如果将前肢的原基去除,支配它的感觉神经节减小,而如果通过移植在胚胎一侧多加一个前肢原基,支配的感觉神经节也会增大。汉伯格1932年到Lillie实验室后重新捡起Shorey的实验,1934年他发表文章确定可以重复Shorey的结果,缺失靶组织后,导致支配它的感觉和运动神经中心减小。1939年他发表文章,说明用鸡胚重复了Detwiler的实验,发现实验增加靶组织会增大支配它的感觉和运动中心。这些研究显示:靶组织调节神经的发育,使支配的神经和靶组织之间相匹配。

为什么神经与其外周靶组织会匹配?Detwiler提出靶组织调节神经细胞的分裂和增殖,汉伯格提出靶组织发出的信号,来影响未分化的神经细胞进行分化。1944年汉伯格和Keefe实验证明,细胞增殖并不参与此过程。

汉伯格将1934的文章的单行本寄给意大利的Levi教授。Levi将论文给了学生蕾塔·列维-芒塔齐尼。蕾塔于1981年回顾:“那是墨索里尼对法宣战的1940年6月,有一天我在送牛的火车箱地板上,读Viktor的文章”。Cattle coach是用于运送动物的货车车厢,有时用来载人,失业的蕾塔在牛车上也急于了解科学发现。

蕾塔虽然看重汉伯格的结果,但不同意其解释。她没有实验室,就在卧室做研究,到菜市买鸡蛋,做完实验把剩下的鸡蛋吃了。她和Levi的文章发表于1942的《比利时生物学志》。她重复了汉伯格的实验,但更仔细地分析了神经节内的神经细胞数量。她看到,在胚胎发育的特定几天,未分化的神经细胞数量随发育而减少,因为分化了而长出神经纤维的神经细胞数量在同一阶段增加。她发现,如果去除鸡胚一侧后肢的肢芽,未分化的神经细胞数量不受影响(等同于未去除肢芽的一侧),而分化的神经细胞数量逐渐减少。她认为这些结果提示:去除肢芽不影响神经细胞的分裂和分化,而影响神经细胞的生存。

战后,汉伯格读到蕾塔和Levi的文章,1946年他邀请蕾塔来他实验室,不同意见的研究者开始合作。1947年秋天蕾塔到圣路易斯计划做半年实验,结果待了近三十年。1949年,汉伯格与蕾塔·列维-芒塔齐尼合作发表论文,实验是蕾塔做的,分析的是她熟悉的感觉神经节。该研究不仅通过染色可以看到细胞蜕变,证实1942和1944年她的解释是对的,而且发现,在正常发育过程中,自然出现大量的细胞死亡。他们也看到,细胞死亡发生在神经纤维已经长到靶组织的时候。他们提出,去除靶组织导致支配的神经细胞的蜕变,而在正常发育过程中,纤维长到靶组织的神经细胞数量多于靶组织的需要,过多的神经细胞得不到靶组织环境的条件而死亡。同年开会时,汉伯格和蕾塔(1950)用了营养性(trophic)一词,说明靶组织对神经细胞的作用。

至此,确立了几个概念:发育过程中自然发生细胞死亡、靶组织对神经细胞生存的影响、细胞间的营养性作用。

以上工作,汉伯格和蕾塔·列维-芒塔齐尼都起了重要作用。之前,虽然1909和1920年有过实验、但问题的重要性未被认识,汉伯格在1934年后积极推动,确立现象后力图建立概念。而蕾塔·列维-芒塔齐尼首先得到正确的解释,他们合作建立了多个新概念。

15A2神经生长因子

神经生长因子的研究,蕾塔·列维-芒塔齐尼的工作多于汉伯格。

不过,1939年,汉伯格就曾提出:“外周产生控制生长的因子(growth controlling agent),向心性传到运动中心”。这个概念,等于是神经生长因子的影子。但这并非他当时唯一的解释,他和蕾塔1949年文章还提出过两种解释,其中第二个解释与此类似。

汉伯格以前的一个研究生Elmer Bueker(1943,1945)一边打工养家一边做研究生论文,他重复汉伯格的肢芽移植实验,结果相同。1948年,已到外地任教的Bueker别出心裁,把肿瘤移植到鸡胚,他想看,生长快的肿瘤能否影响神经细胞的生长。他首先用小鼠乳腺腺细胞肉瘤(肉瘤180)和Rous禽肉瘤,结果发现移植Rous禽肉瘤的鸡胚都死了,移植肉瘤180的鸡胚常常把肉瘤给吸收了。但是,在没有被吸收的几例中,肉瘤得到了神经支配,而且支配它的感觉神经节体积大于正常。

蕾塔和汉伯格曾试图找可以影响神经细胞生长的组织,发现脑、肝、肌肉和皮肤都不行。汉伯格于1948年秋给蕾塔看Bueker的文章,蕾塔看后决定也做肉瘤。汉伯格写信给Bueker征求他同意重复和扩充其研究,然后从Jackson实验站要了四种肉瘤,肉瘤180以外,还有肉瘤37以及另外两种乳腺瘤。蕾塔重复了Bueker的实验,也移植过老鼠胎盘,结果肉瘤180和37能增加感觉神经节细胞的分裂、分化和生长,老鼠胎盘对鸡胚无此作用。肉瘤的作用不依赖于神经纤维和肉瘤细胞建立突触连接,这些结果由Levi-Montalcini和Hamburger在1951年发表。蕾塔觉得可能有弥散性物质参与此过程,就把肉瘤移植到其它远离神经的地方,结果也有作用,这些发现,她先单独在会议宣读、在会议集发表(1952),1953年与汉伯格共同正式发表。

蕾塔曾试图检验肉瘤的提取物能否有同样作用,没成。然后她想做体外实验。她联系了自己的意大利朋友、当时在巴西工作的Herta Meyer(1902-1990),后者会做体外培养。汉伯格为蕾塔安排洛克菲勒基金会的资助,蕾塔于1952年9月揣了两只移植了肉瘤的老鼠飞到里约热内卢,她把感觉神经节放到体外培养,然后加入小片肉瘤,她把结果画成图寄给汉伯格。1953年1月她回圣路易斯继续同类实验。1954年她和Meyer、汉伯格报道,在体外培养中,她们发现肉瘤180和37在十几、二十小时就可以增加神经节长出很多神经纤维。另外几种瘤没有这样的作用。有作用的肿瘤细胞,只要和神经细胞共培养就行,无需直接接触,说明肉瘤细胞分泌可弥散的分子作用于神经细胞。

如何拿到这种可弥散的分子?蕾塔和汉伯格都没有分离纯化的技术专长。但是,和校本部相隔一个“森林公园”的医学院有。1940和1950年代,正是华盛顿大学生物化学的鼎盛时期。生物化学系以Carl和Gerty Cori夫妇(1896-1984; 1896-1057)为首,他们两人实验室出了8位诺贝尔奖获得者,而微生物系也是生化学家领导,系主任是犹太裔Arthur Kornberg (1918-2007)。他们在华盛顿大学医学院的基础和临床系科形成了很强的生化圈。通过熟悉生化圈,汉伯格邀请在放射科做博士后的Stanley Cohen到校本部汉伯格自己主持的动物系,加入寻找生长因子的研究。雇到Cohen时,蕾塔还在巴西,她回信给汉伯格说“从你的描述看来,他是着手从事发现肉瘤释放分子这样困难问题的正确人选”。

Cohen熟悉生物化学技术,他加入后分离纯化因子的进展很快。1954年,Cohen、Levi-Montalcini和Hamburger通过汉伯格投稿《美国科学院院刊》,报道他们将肉瘤细胞破碎成匀浆,仍可检测到活性,刺激感觉神经细胞长出神经纤维。活性组分在当时未完全分离纯化的情况下,还含核酸和蛋白质,加热可以失活。他们论文的题目是“肉瘤37和180中的神经生长刺激因子”。

此后,汉伯格淡出神经生长因子的研究,蕾塔·列维-芒塔齐尼和Cohen继续。

Cohen不仅在校本部做神经发育的实验,而且经常穿过公园到医学院参加生物化学的学术活动。当时,Kornberg主持的微生物系每天中午有学术报告和午餐,听众中不仅有当时的生物化学大家、也有以后成名的分子生物学大家,如Paul Berg(1926-2023)、David Hogness(1925-2019)、Dale Kaiser(1927-2020),以后都被Kornberg带到斯坦福大学建立生化系。Cohen在微生物系的学术活动中讲了自己的结果,Kornberg说你这是在研究病毒,如果是病毒,里面就有核酸,Kornberg建议Cohen用磷酸二酯酶降解病毒的核酸,来证明是否确有病毒参与。微生物系日裔助理教授早石修(Osamu Hayaishi,1920-2015)用一种蛇毒制备了磷酸二酯酶的粗提物。Cohen和蕾塔赶紧用它做了实验,结果发现,不仅肉瘤的活性没有下降,而蛇毒制备物本身就有很强的活性,反而刺激了神经纤维的生长。原因是这种粗提物不是很纯,不仅有磷酸二酯酶,而且还有生长因子。Cohen和蕾塔1956年发表论文“蛇毒中的神经生长刺激因子”。1957年,他们有更多证据提示此因子是蛋白质,包括它对热敏感、对多种蛋白酶敏感。1960年,Cohen报道小鼠唾液腺中也有神经生长促进作用,用多种方法分离纯化后活性更高,分子量估计4万4千kD,文内用了“神经生长因子”一词。NGF的活性不仅对热敏感、而且可以被蛋白酶所降解。Cohen制备了NGF的抗体,而注射此抗体到新生或成年小鼠体内后,可以破坏交感神经系统。同期杂志中,Levi-Montalcini与Booker发表一篇文章报道Cohen提取的因子也刺激体外培养的人胚胎交感神经节,如果注射到新生或者成年小鼠,也刺激体内交感神经节生长。她们还发表一篇文章,发现注射因子的抗体后,小鼠、大鼠、兔、猫、猴的交感神经都大大减小,仔细分析了其中变化。所以,体外和体内实验证明因子可以促进神经生长,而用功能阻断的抗体可以破坏体内已经生长的神经,证明其对神经维持的必要性。

Cohen的以上工作都在华盛顿大学进行,发表时他已到Vanderbilt大学任助理教授,因为汉伯格系里缺乏经费留Cohen任教。此后,Cohen研究NGF不多,而主要集中研究表皮生长因子EGF。

1960年代,几个实验室试图确定纯化的NGF蛋白质的氨基酸序列,1971年,Angeletti和Bradshaw确定了NGF是由118个氨基酸组成的蛋白质,分子量为13250道尔顿。

汉伯格在1970和1980年代,重新回到神经细胞死亡的研究,他和其他人证明神经生长因子可以抑制正常发育过程中出现的神经细胞死亡。

15A3研究的长处和短处

汉伯格、Levi-Montalcini、Cohen以及后继者,从1950年代初期到1971年寻找NGF的过程,很重要的长处是他们开创一个领域。

在50年代的研究中,他们建立了比较方便的体外实验,也一直注意做体内实验。拿到NGF以后,不仅确定其能对神经细胞发生作用,而且很快就制备了抑制其功能的抗体,而注射抗体可以破坏体内神经系统,证明NGF、或有NGF样免疫特征(对所制备抗体有反应)的分子在体内参与维持神经系统,两套实验分别验证了充分性和必要性。

复习的1950和1960年代工作也可以看到不足之处。他们在做实验过程中换了生长因子来源的靶组织,从最初的肢芽,到肉瘤和唾液腺。那么就有可能:这些不同的组织和细胞含不同因子,肉瘤和唾液的分子可能完全不同于肢芽的分子。用来检测的神经组织也有变化,开始做鸡胚实验是检测感觉神经节中的感觉神经元和脊髓腹角的运动神经元,后来常用交感神经节中的交感神经元。最大的问题,是在检测过程中,他们做过不同的细胞生物学现象,如分裂、分化和死亡。而很有可能,某个分子只有一个作用,而不是所有作用,如果一定要找抑制死亡的分子,那么就要坚持检测死亡,而不是看到长了很多神经纤维就算有活性。刺激神经纤维的生长,可能是细胞分裂多了,也可能是细胞分化多了所以长出很多纤维,也可能是细胞死亡少了。Levi-Montalcini和Cohen在研究过程中,有时注意了用多种检测,有时没有,因为有些检测很快就很方便地得到很强的结果。

用抗体做的实验,为了操作方便,注射到新生和成年小鼠,而用鸡胚做的实验用的发育阶段远远早于1960年抗体实验用的阶段,完全可能NGF只参与早期的发育,而不参与出生后的发育和维持。开始不易用胚胎做抗体注射的实验,以后得到改善,当然最清晰的实验需要等有基因剔除技术后。

40年代观察到肢芽对感觉神经元和运动神经元的作用。1960和1970年代找到的NGF不能解释肢芽的作用,至少不能解释肢芽的全部作用,因为虽然NGF对背根神经节的感觉神经元有营养性作用,NGF对运动神经元没有营养作用。后来发现的其它神经营养性因子及其它生长因子,对运动神经元有营养性作用。现在还有人认为肢芽(和作为靶组织的肌肉细胞)含有其它未知分子,尚需研究。

既然他们没有找到解释肢芽活性的分子,而还有其它一些问题,那么,为什么还是公认他们做出了重要的贡献?

最重要的是,当时很少人研究细胞分裂、分化和死亡,他们无论得到影响其中哪一个现象的分子,都是重要的,这是不做热门而开辟一个领域的好处,如果是在一个研究人员很拥挤的领域,混淆这些问题就不容易进展。

另外,后来知道NGF本身有多种作用。比如,NGF可以刺激神经细胞分化长出神经纤维,这个作用与细胞死亡的作用是分开的,有些其他分子只有刺激分化的作用,不能抑制死亡。NGF两个作用都有。直到1970和1980年代,才有更好的实验证明纯化的NGF直接抑制细胞死亡,而不是通过增加细胞分离、或细胞分化。

15A4越走越宽

1982年,德国的Barde和Thoenen发现脑中神经营养性因子活性BDNF,并于1989年获得其基因,BDNF和NGF是同一个家族,结束了NGF孤单的历史,此后很快多个实验室用分子生物学技术找到多个神经营养性因子,他们分别和协同对不同神经细胞起营养性作用。

NGF是如何起作用的?1986年,美国的Moses Chao获得第一个NGF受体的基因,此受体现在一般称为p75。1991年,两个实验室发现Trk癌基因编码的蛋白激酶也是NGF的受体。Trk与p75有协同有分工,三个Trk分别参与将细胞外不同的神经营养性因子的信号转导到细胞内。

从此,很多人加入研究神经营养性因子的领域,发现更多有趣的特性和功能,比如这些蛋白质从前体加工而来、但“前体”有不同于成熟蛋白质的功能。神经营养性因子的作用,也不止神经细胞的生长和存活…。蕾塔2012年的文章报道用NGF的抗体注射到早期鸡胚后,胚胎的轴转向受到影响,这是早于神经发育的一个步骤。

神经营养性因子,最初十几年是只有几个人做的课题,现在是成百上千神经生物学研究者的领域。而发现细胞的生长因子,有更广泛的意义,存在于针对多种细胞的不同生长因子,除了NGF、EGF,还有FGF、TGF、PDGF、VEGF…遍布生物医学很多领域,而且作用很重要,现在可能上万人的研究和生长因子有关。

无论生长因子、还是神经营养性因子,后来都有很多活跃的研究。

15A5    基础和应用的曲折关系

NGF和EGF的工作,都是基础研究,不是为了应用目的而开始。1980年代以后有很多人尝试,神经营养性因子对疾病中的细胞蜕变是否有治疗效果,很长时间并无这方面的临床应用。

但是,另外一个研究却和应用关系很大。不过,也非当初目的,且无可预见。就是表皮生长因子EGF的研究。

Cohen在离开华盛顿大学以前,偶然发现小鼠颌下腺有生物活性,可以提前让新生小鼠眼睛睁开。他到Vanderbilt大学后继续此项研究,1962年发表第一篇文章,以后纯化此分子,更进一步确定其生物活性,1965年,他命名为EGF。对EGF的研究,特别是其受体的研究,建立了受体磷酸酶的典型。EGF的工作起初只与发育有关,后来其他人发现与肿瘤有关,EGF的受体基因突变后,可以造成肿瘤,其机理相似于Rous肉瘤产生的原理:都是因为蛋白激酶活性过高造成。

二十年后发明针对EGF受体的药物,EGF受体选择性拮抗剂,是通过分子机理来进行合理药物设计的典范,成为今天抗肿瘤的常用药物。比较可笑的是,Cohen和蕾塔当初用肉瘤做NGF,最后却与肿瘤关系很小,而Cohen用老鼠睁开眼皮的实验,却与肿瘤关系很大,不过不是眼皮的癌症,而是肺癌等其他常见癌症。生物学研究中,有时并非很容易从最初用的材料来确定是否将来有实用价值。
